PCR process:

The PCR process can be roughly divided into three steps: denaturation - annealing - extension.

1. Denaturation (90ºC-96ºC) : Double stranded DNA template under heat, hydrogen bond breaks, forming single stranded DNA.

2. Annealing: (60ºC-65ºC) : the system temperature decreases and the primer binds to the DNA template to form a local double strand.

 

3. Elongation: (70ºC-75ºC) : Under the action of Taq enzyme (about 72ºC, the best activity), using dNTP as raw material, starting from the 3 'end of the primer and extending from 5' to 3 'end, the DNA chain complementary to the template is synthesized.

As shown in above, after a denaturing annealing process, the original DNA double strand was changed into 2pcs double strands. Dozens of cycles can produce a large number of identical DNA fragments.
